The metabolic processing and cellular localization of 2,4-diaminotoluene was studied in the mouse. In vivo metabolism of labeled 2,4-diaminotoluene led to the formation of a major metabolite in the urine and all major metabolites in the liver. Two metabolites formed in the liver, a polar and a neutral, contained one dihydroxy (OH) and one amino (NH2) group, respectively. Both were identified by mass spectrometry. In addition, the mercapturic acid conjugate of 2,4-diaminotoluene was identified in the urine after the administration of 14C-labeled 2,4-diaminotoluene to mice. The time course of excretion of the mercapturic acid conjugate was slower than that of the mercapturic acid metabolite of benzo[a]pyrene and differed from the time course of excretion of the mercapturic acid metabolites of the fungicide vinclozolin. Cytosolic and microsomal subcellular fractions isolated from mouse liver were used to identify cellular compartments of the route of processing of 2,4-diaminotoluene. These experiments demonstrated that the mercapturic acid conjugate of 2,4-diaminotoluene was metabolized by the cytosol. Our data are the first to demonstrate that the mercapturic acid conjugate is formed in a microsomal system in vivo and that the cytosolic system is responsible for the metabolism of 2,4-diaminotoluene.
